060 Influence of low- intensity electric field on the interaction of some microorganisms with silver and copper ions |
Authors: | PodolskaV.I., Voitenko Ye.Yu., Yakubenko L.N., Ulberg Z.R.,
Summary: | The peculiarities of weak pulsed electric field influence on metal ions bioaccumulation and colloid silver/copper formation in bacteria and yeast cells were investigated. Bioaccumulation value increased as a result of superposition of biosorption and field induced sorption. Amplification of bactericide properties of hybrid materials based on microorganisms and silver (copper) nanoparticles as a result of synergetic effect of copper additives, which catalyzed mixed silver and copper particles formation, was observed.

Authors: | Nikovskaya G.N., Godinchuk N.V., Samchenko Yu.M.
Summary: | The results of the removal of Cu (II) and Fe (III) heavy metals from individual, mixed solutions and natural water by means of hydrogels sorption are provided. Copolymer acrylic hydrogels and agar-agar were used. It was found that sorption of heavy metals occurs synchronously with the hydrogels swelling. The efficiency of heavy metals removal (up to 99% for individual solutions with the 2 mM concentration, and up to 95% for natural water) and strength of its retention are maximum for copolymers of acrylamide and acrylic acid.

060 Bioextraction of heavy metals from colloidal sludge systems |
Authors: | Kalinichenko K.V., Nikovskaya G.N., and Ulberg Z.R.
Summary: | Chemical (acidic) and biological (under the action of acidogenic metabolites synthesized by microorganisms present in a sediment) extraction of heavy metals from colloidal sludge sediments produced at a waste water biochemical treatment plant is studied. It is established that both methods provide at least 80% extraction of heavy metals, which decreases in the sequence Zn > Mn > Cu > Ni > Co > Pb > Cr. The advantage of this biological procedure is the possibility of using the processed sludge sediments, which contain many useful components, as fertilizers.

060 Changes in the surface properties and stability of biocolloids of a sludge system upon extraction of heavy metals |
Authors: | Kalinichenko K.V., Nikovskaya G.N., and Ulberg Z.R.
Summary: | The influence of chemical and biological extraction of heavy metals on the stability of natural sludge sol, as well as on the wetting and electrosurface properties of its particles, is studied. The biological extraction of metals is found to result in hydrophobization of sludge colloids and reduce their negative charge. Heavy metal extraction destabilizes the sludge sol, the level of destabilization being maximum upon bioleaching. The presence of exopolysaccharides considerably stabilizes the sludge system; however, it weakly affects the system’s stability after extraction of heavy metals.

060 Sediments of municipal waste water. Biotechnologies of utilization |
Authors: | Nikovskaya G.N., Kalinichenko K.V.
Summary: | Actual ecological and chemical problem of rational utilization of sludge waste, which are generated as a result of the widespread biological treatment of municipal wastewater, is discussed in the review. Hundreds millions tons of the sludge solids per year are produced and occupy vast suburban areas. Information on the spread, toxicity, biochemistry, microbiology, colloidal and chemical properties of the sludges is represented. The municipal sludge wastes are highly concentrated, heterodisperse biocolloidal system stabilized by biogenous high-molecular compounds. These include microbial cells, their metabolites, products of organic and mineral contaminants transformation and heavy metals compounds as main components. Due to the presence of micro- and macro-elements, vitamins, amino acids, beneficial microorganisms and organic matter, the most rational approach to the utilization of the sludge solids is their use as an agricultural fertilizer after partial removal of toxic heavy metals. Therefore, the interaction of sludge components with heavy metals, modern biotechnologies of their removing from natural colloidal systems are discussed. These technologies based on the enhancing of vital ability of sludge autochthonous biocenoses by adding essential nutrients and synthesis of metabolites with the properties of flocculants and heavy metals extractants. The possibility of the municipal sludge wastes conversion into fertilizer by starting the biological processes with heterotrophic sludge microorganisms at middle and high temperatures are described.

060 Biotransformation of sludges of municipal wastewater biological treatment into fertilizer |
Authors: | Nikovskaya G.N., Kalinichenko K.V.
Summary: | As a result of the biological treatment of municipal wastewater, sewage sludge wastes in the form of high-concentrated suspensions contaminated by heavy metals and stabilized by the treatment under aerobic-anaerobic conditions are produced. The process of the biotransformation of these suspensions into fertilizers has been developed. It is based on the activation of vital ability of the sludge heterotrophic microorganisms by the addition of necessary nutrients, leaching of heavy metals, sludge suspension flocculation and separation to liquid and solid phases. Each of these can be used as a fertilizer after the treatment with potassium hydroxide at .pH 10.0—11.0 and acidity correction up to .pH 8.0 by phosphoric acid. The suggested method for the sludge treatment made it possible to obtain solid and liquid fertilizers with the decreased content of heavy metals (up to the permissive concentrations) and enriched by ecologically valuable microorganisms. The introduction of the fertilizers enhanced the plant biomass productivity by 5—10 times.

060 Chemical-microbiological and biogenous formation of ultrafine silver in lactic acid bacteria cells |
Authors: | V.I. Podolska, O.Yu. Voitenko, N.I. Grishchenko, Z.R. Ulberg, O.G. Savkin, L.M. Yakubenko
Summary: | The aim of this investigation was to study the influence of bacteria-matrices and the method of silver nanoparticles synthesis on the formation of hybrid materials based on the Lactobacillus cells separated from the known products and preparation. These cells were used as natural templates for silver nanoparticles precipitation due to a presence of supramolecular layer on the Lactobacillus surface with the pores of 2-8 nm size. Hybrid materials based on Lactococcus lactіs, Lactobacillus acidophilus and Lactobacillus plantarum cultures were synthesized by a chemical-microbiological method of sorption reduction from ammonium silver complex solution using the reducing agent of sodium borohydride and by a biogenic reduction method with active groups of a cellular wall into an alkaline dark environment. The silver ultrafine particles formation controlled by the method of spectroscopy in ultraviolet-visible areas. The presence of silver particles was confirmed by means of energy-dispersive X-ray spectroscopy. The particles size distribution was estimated from TEM micrographs of cells with silver nanoparticles using the ImageG program. The differences in physiology and surface structure properties of investigated cultures of Lactobacillus as well as the methods of silver nanoparticles formation affected the structural characteristics of silver particles and spectral properties of the hybrid materials notably. The biogenous formed hydride materials had the yellowish brown colouring, materials formed by chemical-microbiological method were dark-brown coloured. We observed that plasmon resonance band of the biogenic silver nanopartricles occurs at 380-400 nm. Particles formed by chemical-microbiological method demonstrated a significant shift in the peak wavelength to 440-460 nm and shoulder at 400-440 nm. Apparently, the biogenic silver nanoparticles have a less size and achieve improvement in the monodispersity comparatively with particles precipitated in bacteria cell by sodium borohydride. Treatment of Lactobacillus biomass by an external low pulse electric field ( 20V, 2,0 KHz, 100 sec ) at the time of silver ions accumulation (30 min), as well as disintegration of surface S-layer lattice of a bacterial cell by guanidine hydrochloride or by acidic hydrolysis caused the shift of plasmon resonance band to the red region and additional peak appearance. Such changes can be related to a wide size distribution of the particles with different form of aggregates formed in the cell wall. L. plantarum and L. acidophilus lactobacillus were more effective for the formation of ultrafine silver particles with biogenic and chemical-microbiological methods in comparison with L. lactis culture.

Summary: | This work reports the preparation and characterization of silver nanoparticles synthesized intracellular through biogenic method and chemical-microbiological method of reductive sorption in Lactobacillus plantarum and Lactobacillus acidophilus cultures when the whole cells were used for nanoparticles production. The hybrid materials based on bacteria cells were synthesized using the reducing agent of sodium borohydride and active groups of a cellular wall. The biogenic formed hydride materials had a yellow-brown coloring, the materials formed by chemical-microbiological method were dark-brown colored. The methods of TEM microdiffraction, dispersive-X-ray-fluorescence analysis and transmission electron microscopy have been used to characterize the prepared silver nanoparticles in such hybrid materials. The both methods used allow precipitating the small mainly spherical subcolloid particles with relatively narrow size distribution. Biogenic produced particles were more uniform in size, 90% of them have an average size of 4 nm. Silver particles precipitated with reduced agent of sodium borohydride were also ultrafine. More than 70% of them fall in the range of 3–5 nm. It was estimated that the biological matrix of microorganism cells, used for nanoscale silver formation, especially surface S-layers, affects their phase and disperse characteristics. Functionalization of silver particle surface with organic ligands such as proteins, polysaccharides, or polyols adds some features to the nanoparticles. It has an influence not only on size, but crystallographic parameters as well. The interplanar distances determined from microdiffraction pattern corresponds mainly the cubic structure of crystals. A bit conservative values for the index face (111) and (220) indicate the presence of strain in these ligand-stabilized crystals of very small size. The data obtained may be useful for the development of new hybrid materials based on cells impregnated with ultrafine metallic and inorganic particles.
